Resolution criteria

Gap closes when: (1) A 12-week chronic TFEB overactivation study in wild-type mice (via AAV-TFEB or doxycycline-inducible transgene) measures lysosomal biogenesis markers, mTOR activity, and neuronal viability by monthly timepoints; and (2) the study reports no lysosomal storage accumulation (assessed by LysoTracker and EM ultrastructure) and no significant neuronal loss (<= 10% reduction in NeuN+ cells) at the effective dose; and (3) transcriptomic profiling (RNA-seq) at endpoint shows sustained autophagy upregulation without dysregulation of apoptotic pathways (< 2-fold change in caspase-3/9 at FDR < 0.05).
