It remains unresolved whether mature oligodendrocyte (MOL/MFOL) depletion precedes or follows expansion of MGnD/DAM microglia (marked by SPP1, GPNMB, LPL, APOE) across the Braak staging spectrum in aged human cortex. SEA-AD snRNA-seq data span Braak I–VI in ~84 donors, making this a tractable compositional regression question, but no published analysis has formally tested the temporal ordering using Dirichlet regression or scCODA with Braak as a continuous covariate in both lineages simultaneously.
