PMID:39733844 discussed that the APP A673V mutation (A2V) is protective against amyloid pathology while A2T promotes aggregation. The structural mechanisms determining how these adjacent substitutions in Abeta have opposing effects on aggregation kinetics and toxicity are not fully understood. The peptide Aβ1-6A2V(D) shows neuroprotection but the therapeutic window and mechanism need elucidation.
