PMID:10.1101/2025.04.28.651121 identified the Neu1-PPCA lysosomal complex as a critical regulator of APP sialylation and processing, with Neu1 deficiency enhancing amyloidogenic cleavage. The mechanisms by which Neu1/PPCA deficiency leads to enhanced amyloid-beta production, and whether restoring Neu1 activity is therapeutically feasible in AD are critical next steps.
