Resolution criteria

Resolved when an evidence artifact resolves the mechanistic basis for myelin-specific T cell dual function (oligodendrogenesis versus demyelination) with one of: (1) single-cell RNA-seq of T cells infiltrating MS lesions or cuprizone demyelination models, identifying >=2 distinct myelin-specific T cell clonal lineages with differential cytokine profiles (Th1/Th17 versus regulatory IL-10-producing subtypes), linked to their oligodendrogenic versus destructive effects; (2) antigen specificity studies showing that myelin basic protein (MBP)-specific versus myelin oligodendrocyte glycoprotein (MOG)-specific T cells have opposing effects on oligodendrocyte precursor cell (OPC) differentiation in vitro, with ELISPOT or single-cell RNA-seq confirming cytokine profiles; (3) adoptive transfer experiments in cuprizone demyelination models demonstrating that IL-10-deficient myelin-specific T cells fail to promote oligodendrogenesis while WT T cells do (>=2-fold increase in CC1+ OLIG2+ oligodendrocytes), establishing IL-10 dependency. Quantitative threshold: n >= 6 animals per group, >=30% difference in OPC differentiation.
