Description

The abstract describes phagocytosis having dual roles but doesn’t explain the mechanistic switch between protective and harmful functions. Understanding this transition is critical for therapeutic targeting without disrupting beneficial microglial activities.

Gap type: unexplained_observation Source paper: Microglia at the Forefront: New Insights From the Glial Club South Cone Meeting 2025. (2026, J Neurochem, PMID:41668347)

Resolution criteria

Resolution requires: (1) Single-cell RNA-seq of microglia sorted from AD/PD mouse models at disease stages identifies transcriptional signatures distinguishing repair-state (DAM1, homeostatic overlap, Trem2-high) from detrimental-state (DAM2, pro-inflammatory) phagocytes, with >=2-fold differential expression in >=50 genes (FDR<0.05); (2) Optogenetic or chemogenetic manipulation of microglia at specified disease stages in vivo shows that enhancing phagocytosis early (pre-plaque) reduces pathology >=25% while enhancing at late stages worsens neuronal loss >=20%, establishing stage-dependent directionality; (3) Identification of >=1 molecular switch (lipid sensor, damage signal, or cytokine) whose genetic or pharmacological targeting shifts microglia from detrimental to beneficial state in vivo with measurable rescue of neuronal viability (>=30% preservation). In vitro phagocytosis assays without in vivo stage-specific manipulation are insufficient.
