Resolution criteria

Resolution requires: (1) In vitro fibril kinetics: ThT fluorescence assay showing SARS-CoV-2 proteins (nucleocapsid, spike fragments, or nsp) accelerate Abeta42 or alpha-synuclein fibril formation with >=3-fold faster t50 at equimolar concentrations, with dose-response across >=4 protein concentrations; (2) Cryo-EM or AFM structure of co-aggregates identifies specific interaction interfaces, with alanine-scanning mutagenesis confirming >=2 residues essential for co-aggregation acceleration; (3) Cellular validation: neurons or iPSC-derived dopaminergic cells exposed to SARS-CoV-2 protein show >=2-fold increase in intracellular alpha-syn inclusions (pS129+ puncta by immunofluorescence) and >=25% reduction in viability, reversible by peptide inhibitors targeting identified interface. Epidemiological association between COVID-19 and neurodegeneration risk without mechanistic data is insufficient.
