Brain swelling during neuroinflammation creates mechanical stress that activates astrocytic Piezo1 channels, amplifying their inflammatory response. Selective Piezo1 inhibition could break this mechanical-inflammatory feedback loop, preventing edema-driven escalation of the microglia-astrocyte cycle while preserving normal mechanosensation.
