What resolves this contention: Polarity and segregation of dSPN function: classical model assigns dSPNs a uniform motor-promoting role with direct-pathway monopoly, whereas dorsolateral dSPN stimulation suppresses movement and asymmetric inputs implicate D2-SPNs in driving direct-pathway elements. / In contrast to the canonical motor-promoting response of dSPNs in the dorsomedial striatum (DMSdSPNs), optogenetic stimulation of dSPNs in the dorsolateral striatum (DLSdSPNs) suppressed locomotion. / These results are consistent with the functional connectivity in which the excitatory inputs to D1-SPNs mostly drive the direct pathway, and the inputs to D2-SPNs target both the indirect and direct pathways ( Figure 3A ).
