What resolves this contention: Disagreement on the relative quantitative weight of ascending L5-IT→L2/3 excitation versus the canonical L4→L2/3 input; partly attributable to different methods (paired patch vs 2P opto) and different cortical areas (S1 vs V1). / In mouse V1, two-photon optogenetics finds strong excitatory connections from L5 intratelencephalic neurons onto L2/3 pyramidal cells — adding a prominent ascending L5→L2/3 path to the canonical microcircuit. / In mouse C2 barrel column paired recordings, the strongest excitatory influence on the column is provided by presynaptic L4 neurons; ascending L5→L2/3 connections are quantitatively much weaker than L4→L2/3 in the layer-pair P(conn) matrix.
